Vibration-induced white finger disease (VWF) is caused by continued use of vibrating hand held machinery (high frequency vibration >50 Hz), and affects tens of thousands of people. New research published in BioMed Central's open access journal Clinical Epigenetics finds that people with a genetic polymorphism (A2191G) in sirtuin1 (SIRT1), a protein involved in the regulation of endothelial NOS (eNOS), are more likely to suffer from vibration-induced white finger disease.
VWF (also known as hand arm vibration syndrome (HAVS)) is a secondary form of Raynaud's disease involving the blood vessels and nerves of arms, fingers and hands. Affected fingers feel stiff and cold and loose sensation for the duration of the attack, which can be very painful. Loss of sensation can make it difficult to carry out manual activities. Initially attacks are triggered by cold temperatures but as the disease progresses attacks can occur at any time.
Little is known about what causes the restriction in blood flow, however researchers from Germany investigated the role of SIRT 1 by looking at polymorphisms (naturally occurring variations in DNA sequence) in people affected by VWF.
SIRT1 regulates activation of other genes by controlling how tightly DNA is wound in the nucleus. Tightly wound DNA cannot be 'read' and consequently cannot be used to make new protein. SIRT1 is known to regulate vasodilation by targeting eNOS, a nitric oxide synthase within the cells lining the inside of blood vessels, which regulates smooth muscle contraction, and hence the diameter of the vessel, and the amount of blood that can flow through it.
Of 113 polymorphisms tested, in the gene coding for SIRT1, only four actually affected the protein, the rest were non-coding or false positives. Of these four, only one was different between people with VWF and unaffected controls. A single nucleotide at position 2191 can either be an A or a G. In the unaffected population 99.7% had the A, but amongst the patients with VWF, almost a third had the G.
Dr Susanne Voelter-Mahlknecht from the University of Tuebingen, who led this study, explained, "While this does not mean that only people with the G version of the gene for SIRT1 will get VWF, it can be used to identify a set of people who would be at risk of VWF if they used vibrating hand held tools. Testing for this variant before starting to work with vibrating machinery could prevent years of pain and disability."

Wesson Ashford, MD, Ph.D. A major goal will be to determine whether treatment with leptin reduced the levels of a protein called tau, which is elevated in the cerebrospinal fluid of Alzheimer patients. Replacement therapy in leptin continued .High levels of the hormone leptin regulation of energy was associated with low rates of disease in a study published in TheJournal of the American Medical Association.
Leptin has been shown to reduce the levels of B-amyloid, the main constituent of the deposits, or plaques, that occur in the brain of people suffering from Alzheimer?s disease.
If our results are confirmed by others, leptin levels in older adults may serve as one of many possible biomarker for healthy aging and the brain, especially, may open new avenues in order to possible preventive and therapeutic interventions, write the researchers. Leptin replacement therapy
But Seshadri said it remains to be seen whether leptin replacement therapy would benefit humans with Alzheimer?s or help protect against disease.
He said the study reinforces the growing awareness that the health problems associated with procedures such as obesity and resistance increase the risk of dementia in old age.
Tau protein is a key that allows the transport along the fibers of the brain, he tells WebMD. If leptin decreases the levels of tau protein in cerebrospinal fluid, which tells us something is happening. So we only need 45 patients to demonstrate an effect.
Over 12 years of follow-up, those with lower levels of leptin were about four times more likely to develop Alzheimer?s than those with the highest levels.

The treatment with leptin has been shown in studies on animals recent to improve the performance of the memory, even after the onset of Alzheimer?s disease, such as dementia.
Discovered in the mid-1990, leptin is produced by fat cells and is considered critical for the regulation and. But there is evidence suggesting a role more and more of the hormone in brain development and memory.
Higher concentrations at the beginning of leptin were associated with lower rates of dementia years later. This association was observed even after adjusting for the impact of quarantine abdominal or belly fat, which was recently identified as a risk factor for Alzheimer?s disease early.
The study initially included 785 elderly participants in the Framingham Heart Study in progress, which began enrolling patients in 1948.
The National Institutes of Health recently awarded nearly $ 3 million for a small pilot study of leptin replacement therapy in patients with Alzheimer?s disease.
?Our skeleton with its bones, joints and muscle connections serves us so well in our daily lives that we hardly pay attention to this great system,? said Dr. Elazar Zelzer Department of Molecular Genetics at the Weizmann Institute. ?While previous research has uncovered the mechanisms that contribute to the development and growth of each component of this complex organ and wonderfully adaptable, specific interactions between bones, muscles and tendons that drive the entire musculoskeletal system are not fully understood.

We couldn?t live without our immune systems, always tuned to detect and eradicate invading pathogens and particles. But sometimes the immune response goes overboard, triggering autoimmune diseases like lupus, asthma or inflammatory bowel disease.
A new study led by University of Pennsylvania researchers has now identified a crucial signaling molecule involved in counterbalancing the immune system attack.
?The immune response is like driving a car,? said Christopher Hunter, professor and chair in the Department of Pathobiology in Penn?s School of Veterinary Medicine. ?You hit the accelerator and develop this response that?s required to protect you from a pathogen, but, unless you have a brake to guide the response, then you?ll just careen off the road and die because you can?t control the speed of the response.?
The research to characterize this immune system ?brake? was led by Hunter and Aisling O?Hara Hall, a doctoral candidate in the Immunology Graduate Group. Additional Penn collaborators included scientists from the Penn Genome Frontiers Institute?s Department of Biology and the Perelman School of Medicine?s Department of Medicine. Researchers from Merck Research Laboratories, the National Institute of Allergy and Infectious Disease, Harvard Medical School and Janssen Research and Development also contributed to the work, which was published in the journal Immunity.
"Healthy people have these cells - you have them, I have them - that are called Tregs,? or regulatory T cells, Hunter said. ?If you don?t have them you develop spontaneous inflammation and disease.?
Different forms of regulatory T cells operate as the brakes on various kinds of inflammation, but, until now, scientists hadn?t been certain of how these Tregs became specialized to do their particular jobs.
Hall, Hunter and colleagues decided to follow up on a molecule called IL-27. Scientists used to think IL-27 played a role in causing inflammation, but, in 2005, a team of Penn researchers, including Hunter, found the opposite; it was actually involved in suppressing inflammation. Thus, when mice that lack IL-27 are challenged with the parasite Toxoplasma gondii, they develop overwhelming inflammation.
?We never worked out how it did that, but it was a paradigm change at the time,? Hunter said.
In the new study, the researchers delved deeper into IL-27?s role. They found that exposing regulatory T cells to IL-27 promoted their ability to suppress a particular type of inflammation. The Penn-led team also demonstrated that they could rescue infected IL-27-deficient mice by giving them a transfusion of regulatory T cells. This finding suggests that IL-27 is required to produce the Treg cells that normally keep inflammatory responses in check during infection.
?Very surprisingly, we were able to show that the Tregs could ameliorate the pathology in this system,? Hall said. ?We don?t think this is the only mechanism by which IL-27 limits immune pathology, but it sheds light on one mechanism by which it could be functioning.?
Further experiments showed that Tregs express a different suite of genes in the presence of IL-27 as compared to another molecule that has been implicated in this process, interferon gamma, or IFN-?. The researchers? findings indicate that the two molecules have division of labor when it comes to suppressing inflammation: IL-27 seems to be important in helping control inflammation at the site of inflammation, whereas IFN-? appears more significant in the peripheral tissues.
?At the site of inflammation, where you?re getting your pathology, that?s where IL- 27 is important,? Hall said.
With a new understanding of how IL-27 may cause a class of Tregs to become specialized inflammation fighters, researchers have a new target for ameliorating the unwanted inflammation associated with all kinds of autoimmune conditions.
?Now we have a molecular signature that may be relevant in inflammatory bowel disease, in multiple sclerosis, in colitis and Crohn?s disease, in rheumatoid arthritis, in lupus,? Hunter said.
Next on tap, the team plans to study IL-27 in the context of asthma, lupus and arthritis.

Being the industry underdog means you're always in need of a punchy pitch. Fortunately, AMD's latest briefing to journalists in London yesterday contained exactly that: for something like the cost of an Intel Core i3 you'll be able to pick up an overclockable Trinity A10-5800K which, we're told, belongs in the same weight class as a Core i5 with HD 4000 graphics. Exact pricing won't be revealed until the full stack of A10, A8, A6 and A4 processors hits shelves next week, but the top-end A10 will likely cost around $130, based on recent leaks and a glance at what Newegg currently charges for an i3. Unluckily, however, whereas Ivy Bridge was compatible with some previous-generation motherboards, Trinity will require the purchase of a new Socket FM2 motherboard.
The claim of performance parity with the Core i5 just cries out to be tested, but we'll have to wait until early October before we can round up verdicts from full reviews on specialist sites. In the meantime, check out the More Coverage links below for some early previews. Also, if you require something more directly head-to-head with an Intel chip, then that's exactly what you'll find in the video after the break, albeit under AMD's auspices and solely in relation to a single game, Sleeping Dogs. As you'll see, there's nothing to turn hardcore gamers against discrete graphics cards, and there are no clues about non-gaming performance (which is arguably more relevant on an APU-powered system). But the A10-5800K does offer plenty of scope for escapism on a low-power HTPC or all-in-one. Indeed, the more expensive Core i5 is left for dust, not least because it's locked -- unlike AMD, Intel charges a premium for its overclockable K-denoted chips. We'll add further preview links as they become available.
Update: Just added HotHardware's preview, which suggests that the A10 really does game as well as our video suggests in addition to providing a mostly fluid computing experience. As mentioned, however, it's impossible to reach a final verdict until AMD allows sites to publish full benchmarks next week.

If you know anything at all about PPC, you know that campaign optimization is one of the most important tasks a PPC manager can perform. Managers spend most of their day tweaking bids, ad copy, campaign settings, networks, keywords, negative keywords, and many other data points within the PPC engines.
Campaign optimization is absolutely essential to PPC success. But if you?re spending all your time in the AdWords and Bing Ads interfaces, you?re missing a big part of the PPC optimization picture.
PPC metrics like CTR, conversion rate, cost per conversion, CPC, etc. are crucial elements that can?t be ignored. This data tells us what is happening with an account: how much traffic it?s generating, how many sales or leads it?s driving, and how much all of that cost. We get a great picture of what is happening.
The problem is, often we don?t know why.
That?s where web analytics come in. Web analytics tell us what PPC visitors did once they arrived at the website.
?Now wait a minute!? you might be thinking. ?I?m using AdWords conversion tracking, so I can see conversions! Don?t those happen on the website??
The answer is absolutely yes. And if you?re not tracking conversions via either the free PPC engine tracking scripts or a third-party tool, then shame on you.
But does that data tell you why someone converted? How many visits to the site did it take for that conversion to happen? What other pages did they view? Were they already a customer making a repeat purchase, or was this their first visit?
PPC conversion tracking can?t answer those questions. But web analytics can.
Even the most rudimentary web analytics measures can tell us something about our PPC campaigns. The following metrics can be found in any web analytics program. I?ll focus on Google Analytics, because it?s so ubiquitous ? but you certainly don?t have to be using Google Analytics to get these numbers.
Bounce rate is the percentage of site visitors who visited one page and then left the site.
Usually, a high bounce rate is considered a bad thing. If you?re an ecommerce site, and 80 percent of your PPC visitors are bouncing, that?s not good ? it means that only 20 percent of people are bothering to go beyond the landing page.
But what if you?re using PPC to generate leads, and you have a lead form right on the page? Visitors could conceivably fill out the form and convert right there, without going to another page. In this case, it?s a low bounce rate that?s bad ? it means no one is converting!
As you look at bounce rate, think about your campaign goals, and what the numbers mean.
Average time on site measures how long visitors spend on your website, in minutes.
It probably takes at least 4-5 minutes to complete an online order on an ecommerce site, so if you?re an ecommerce PPC advertiser, a longer time on site is good.
What about the one-page lead form, though? Best practices for online lead forms indicate that shorter forms are best. If it takes 4-5 minutes to fill out your form, you?re using the wrong form. In this case, shorter times on site are a good thing!
This metric is exactly what it sounds like: the average number of pages visited on your site.
Most ecommerce shopping carts are at least 4-5 pages. Add 1-2 pages for your landing page and any additional items the visitor might be interested in, and you?re looking at a good average of 5-7 pages per visit at a minimum.
I bet you know what I?m going to say here. For the one-page lead form, if your average number of pages visited is 5-7, you?ve probably lost the lead. An average of 1.5 is probably good in this case.

Some partners deal with the fear by driving the desire for a new romance into the deeper recesses of their being, along with the lack of fulfillment that they feel in their current relationship, and they just get busy with other things in life.
There are still others who are engaging in what they believe to be harmless flirtations. They will miss all the signs that indicate they are having an emotional affair by rationalizing that there is no harm in having intimate relationships with people other than one?s spouse or partner.? Or they decide that there can be no harm in having an emotionally intimate relationship with someone other than your romantic partner as long as you and your partner have an intimate bond that is mutually satisfying.

So he is not open and vulnerable with you and does not know the first thing about emotional intimacy.? So what!? You knew that going to the altar.? It was you that thought you were going to change him.? He did not mislead you.? YOU MADE YOUR BED, NOW LAY IN IT.?
Donna is repeatedly reassuring herself that she is not only wrong for feeling romantically unsatisfied, but she has only herself to blame for her situation evolving the way it has. Needing to win the Tour Championship on Sunday to beat out Rory McIlroy for the FedEx Cup, Snedeker came up with three big birdies on the back nine at East Lake to take all the drama out the final day that had been loaded with possibilities.
Snedeker hit his final tee shot into the grandstand left the 18th green and made bogey. By then, it didn't matter. So dominant was his performance that he was the only player from the last five groups who managed to break par, closing with a 2-under 68.
Along the way, he answered any questions about why Davis Love III made him a captain's pick for the Ryder Cup next week at Medinah.

Snedeker won by three shots over Justin Rose (71) in the Tour Championship, his second title of the year, and collected $1.44 million. And he won the $10 million bonus for capturing the FedEx Cup, which comes with a five-year exemption on the PGA Tour.
McIlroy and Tiger Woods, both of whom could have won the FedEx Cup with a victory Sunday, faded early and were never a factor.
Snedeker joins Woods (twice), Vijay Singh, Jim Furyk and Bill Haas as winners of the FedEx Cup in its six-year history.
McIlroy had won the last two playoff events and three of his last four tournaments dating to his eight-shot win in the PGA Championship. He still is virtually a lock to be voted PGA Tour player of the year, but he had to settle for second place in the FedEx Cup.
"I knew what I needed to do," McIlroy said. "I needed to win. Brandt knew what he needed to do. And he played fantastic. He really deserved it."
It was an emotional week in so many ways for Snedeker, already a high-strung personality. His father, Larry, flew in to watch final round at East Lake, only the second tournament he has attended since having a liver transplant last year.
Snedeker stopped by a hospital in Atlanta to visit Tucker Anderson, the son of his swing coach who was critically injured in a recent car accident and remains unable to communicate with words.
"I asked him if he thought I was going to beat Rory McIlroy, and he gave me a wink," Snedeker said.
He beat everybody at East Lake in a convincing fashion. After taking a double bogey on the par-3 sixth when he hit his tee shot into the water, Snedeker responded with a 40-foot birdie putt on the eighth, an 18-foot birdie putt on No. 13, and chipping in from short of the 17th green to end all doubt.
"I had complete confidence in what I was doing," Snedeker said.
He finished on 10-under 270 and will move into the top 10 in the world ranking for the first time in his career.
Ryan Moore made the strongest run at Snedeker on the back nine, and with an up-and-down behind the par-5 15th green for back-to-back birdies, he was tied for the lead. Moore never was in the mix for the $10 million bonus, though he could have affected the outcome of the FedEx Cup.
